Complex assessment of cardioprotective actions of ischemic preconditioning: the influence of ischemia severity on the antiarrhythmic effect

Petrishchev N.N., Tsirlin V.A., Vlasov T.D., Syrensky A.V., Sonin D.L., Minasian S.M., Galagudza M.M.

Abstract

A protective effects of ischemic preconditioning (IPC) on subsequent prolonged myocardial ischemic/reperfusion damage were investigated using both in vivo rat model and isolated heart preparation. Myocardial infarct size, severity and duration of ischemia- and reperfusion-induced arrythmias, and a rate of intraischemic and postischemic contractile recovery were used as an end points of myocardial protection. Accordingly, two major aspects of cardioprotection afforded by IPC could be specified as antiarrythmic and infarct-limiting, i.e., antiischemic. A threshold of antiarrythmic effect of IPC appears to be lower than that of infarct-limiting potency. We failed to see any protection against reperfusion-induced arrythmias when coronary flow was reduced more then two-fold from its initial value. In contrast, IPC confers more early and complete intraischemic contractile recovery in the presence of severe reduction in coronary flow.

Role of opioid and non-opioid systems of the left and right brain hemispheres in formation of ultradian and circadian rhythms of stress-induced hypoalgesia in mice

Kubynin A.N., Mikheyev V.V., Ignatov Y.D.

Abstract

Present work is aimed to study a role of endogenous opioid system in formation of rhythms of stress-induced

hypoalgesia. This problem is investigated from position of functional brain asymmetry. The results of study have demonstrate that phase-shifted circadian rhythm of hypoalgesia is formed as a result of stress-induced activation of endogenous opioid system mostly of right hemisphere structures, as well as opioid response of left hemisphere dominates in generation of 6-h rhythm. The non-opioid mechanisms of antinociception, with dominant influence of right hemisphere, take part in geeation of 8- and 16-h variations of stress-induced hypoalgesia.

Low density lipoprotein subfraction with high negative charge

Denisenko A.D., Silina I.A., Dizhe E.B., Vinogradov A.G., Klueva N.N.

Abstract

A minor electronegative low density lipoprotein subfraction (LDL-) was isolated from human plasma by combination of ultracentrifugation and anionexchange chromatography. LDL represents 5-15% of total LDL. Compared with the major LDL subfraction, designated LDL*, LDL: revealed increased contents of sialic acids. No difference was found in lipid and apoprotein composition between LDL* and LDL. The lipid peroxide concentration evaluated as conjugated dienes content in LDL- was slightly lower, than in LDL*, but there was no difference in susceptibility of LDL* and LDL- to copper-initiated oxidation. LDL' as well as LDL* were taken up by mouse peritoneal macrophages with the same low rate, but uptake of LDL by human lung fibroblasts was 30 % lower as compared to LDL*.Thus, the minor subfraction of LDL with elevated content of sialic acids and lowered affinity to LDL receptor is present in human plasma. It seems reasonable to suppose that elevated negative charge of LDL is provided by high content of sialic acids.

Comparative evaluation of the efficiency of local radiation therapy in the treatment of patients with rheumatoid arthritis

Ignatiev V.K., Marusenko I.M.

Abstract

Conducted evaluation of efficiency of methods of local radiation therapy to which pertain radioactive synovectomy (RS), X-raytherapy (XRT) and method offered by authors - local gammatherapy (LGT). Studied the record a clinic-laboratory factors including immunological under the influence of treatment.

Explored 175 patients suffering rheumatoid arthritis and got local radiation therapy on the knee joints. In the dependence from the type of treatment formed three groups of sick, which were similar on an age, sex and main features of disease. In all groups dominated the women senior 45 years, with duration RA more than 5 years and Il stage on X-ray of disease and moderate degree of general activity. Results: RS exceed other type of local radiation therapy on efficiency, particularly in removed a period of observation. Given method renders anti-inflammatory and immunosuppressional action, as evidenced by the track record of clinical, as laboratory data. Use new method of local radiation action - LGT - allows to obtain positive results more then beside half sick with RA, here with analgetic effect of given type therapy is observed during the path of treatment (62,3%) and exceeded RS LGT beside anti-inflammatory possesses immunosuppressional action herewith noted relaible record of laboratory and immunological data. XRT vastly yields on efficiency to rest methods that connected with the low total dose of irradiation. XRT renders nonspecifical anti-inflammatory action, not infusing on immune inflammation. This is caused smaller and transitory effect of XRT.

Local radiation therapy allows to raise efficiency of drug treatment of RA. Herewith RS and LGT possess immunosuppressional action that ensures their high efficiency, then XRT is characterized by only short-lived antiinflammatory effect.

Concentration of riboflavin in acute alcoholic psychoses

Kirpich I.A., Sidorov P.I., Solovyov A.G., Boiko E.R., Boiko S.G.

Abstract

Under analysis was the dynamics of riboflavin supply of patients' organism with chronic alcoholism in the state of acute alcoholic psychoses. Obvious hypovitaminosis was detected mostly pronounced in patients with somaticaly aggravated psychosis - delirium tremens. А new method is proposed for determining the level of the organism supply with B2 vitamin using riboflavin index calculated bу deviding the index of glutathione reductase enzyme activity bу the index of FAD effect. The quantitative criteria showing the lower limit of the adequate supply of organism with B2 vitamin, the degree of «risk» according to the development of hypovitaminosis and the state of marked hypovitaminosis of organism according to riboflavin .

Metabolic syndrome of insulin resistance and its consequences in endometrial cancer

Berstein L.M., Kvatchevskaja J.O., Gamajunova V.B., Poroshina T.E., Ourmantcheeva A.F., Kovalenko I.G.

Abstract

The signs of insulin resistance syndrome and its association with some features of the disease and DNA damage in somatic and tumor tissue were evaluated in 99 endometrial cancer patients. The frequency of insulin resistance syndrome counted on the basis of fasting plasma glucose and insulin concentrations is equal to 0,35 (95% confidence limits of 0,24 - 0,46) in endometrial cancer patients who do not have a history of diabetes mellitus Patients with well or moderately differentiated endometrial adenocarcinomas have statistically significantly higher basal and stimulated plasma insulin and C-peptide concentrations than did patients with poorly differentiated endometrial adenocarcinomas or rarely encountered tumors of endometrium. At the same time the level of fasting insulinemia positively correlates with disease stage, local and regional tumor spreading in the group of patients with wel or moderately differentiated endometrial adenocarcinomas. On the other side, hyperinsulinemia and other hormonal-metabolic disturbances typical for insulin resistance syndrome do not increase the probability of DNA damage of somatic cells (according to the data of micronucleus test). No association between hormonal-metabolic disturbances and the degree of DNA unwinding in tumor and visually unchanged endometrium has been found too; that is insulin resistance/ hyperinsulinemia (as distinct to hyperestrogenemia) does not result in increasing genotoxical damages in tumor and normal tissue of endometrium. The data obtained confirm once more the necessity of treatment strategies directly aimed at correcting hormonal-metabolic disturbances in endometrial cancer patients.

Andreeva L.I., Boykova A.A., Petrova V.S., Shcherbak S.G., Vorobyov N.V., Belokopytov I.Y., Sarana A.M., Kuzmich V.G., Ivanov I.A., Margulis B.A., Shabanov P.D.

Abstract

Thallium is considered especially toxic metal, its mining and using in industry and yet not long ago in agriculture and medicine connect with accidents of chronic and acute poisoning. Acute food thallium salts poisoning of man   together with specific clinical manifestations leads to pronounced cytotoxical effect. We noted in patient blood normochromic anemia, cytolytical syndrome, a rise of bilirubin, creatinin, cortisole contents. It was found increased capacity of blood mononuclears to generate nitrogen oxide, a high content of tumor necrosis factor a in serum and incubated diluted blood, increased expression of heat shock protein 70 kDa, especially its inducible form. We consider the last fact is a consequence of cell proteins denaturating changes, activation of free radical reactions and is one of the main protection mechanisms to promote survival of ions thallium damaged cells.
